This paper describes a simple, low cost and reliable DNA template preparation protocol for polymerase chain reaction (PCR) using immature leaves from peanut seeds or leaves from field-grown plants. The technique may find wide utility in studies involving PCR-based molecular markers, rapid screening for transformants and gene cloning.

The Latin American peanut industry is estimated using SUR. In scenarios, their demand is not affected dramatically by both price changes. The price changes affect the Latin American supply by roughly 15% and net trade by approximately 50%, compared to less than 10% in world price shock.

Analysis of genomic sequences from peanut (Arachis hypogaea) Electron. J. Biotechnol.

Peanut is an important legume crop across the world. However, in contrast to most legume crops, groundnut lacks taxonomic proximity to any major model genome. A relatively large number of genomic sequences were generated from groundnut as part of a microsatellite marker development project. In the current study, a total of 1312 sequences were analyzed of which 448 contained microsatellite motifs. All sequences (GenBank Accessions: BZ999351-CC000573) were analyzed after clustering for possible similarity with publicly available sequences from Arabidopsis, Lotus, soybean and Medicago. RESUMO: O ensaio objetivou avaliar a adaptabilidade de acessos de Arachis spp. em várzea (solo Glei Pouco Húmico) na Embrapa Cerrados, Planaltina, DF, no período de novembro de 1998 a maio de 2002. O plantio foi realizado com material vegetativo (estolões) em parcelas constituídas de quatro linhas de 2 m de comprimento, espaçadas entre si em 0,5 metro, com área total de 4 metros quadrados e área útil de 1 metro quadrado. O delineamento experimental foi o de blocos ao acaso com quatro repetições. Os tratamentos estudados consistiram de dez acessos de Arachis spp. (BRA 15121, BRA 22683, BRA 30333, BRA 30872, BRA 31496, BRA 31534, BRA 31828, BRA 31801 e BRA 13251). Cloning and analysis of a NBS-LRR disease resistance gene candidate PnAG1 from peanut (Arachis hypogaea L.) Electron. J. Biotechnol.

Background: Based on the conserved sequences of a known NBS resistance gene, a pair of degenerate primers was designed to amplify the NBS-LRR resistance gene from peanut using PCR and RACE methods. Results: Analyzing the amino acid sequence by BLAST on NCBI, which was deduced from the 1088bp-long gene named PnAG1-2, showed that it had a certain homology with some resistance proteins, among which Arachis cardenasii resistance protein gene had the highest homology (66%). Relative quantification PCR analysis indicated that PnAG1-2 gene expresses more in J11 (an A. flavus-resistant variety) than in JH1012 (an A. flavus-susceptible variety) when the harvest time was coming. Effect of gamma irradiation on the inactivation of aflatoxin B1 and fungal flora in peanut BJM

The effect of gamma irradiation on aflatoxin B1 levels and fungal infection were investigated in peanut samples, Tatu Vermelho cultivar. At a radiation dose of 10 KGy, growth of molds was completely inhibited. Doses of 15, 20, 25 and 30 KGy were sufficient for destruction of aflatoxin B1 by 55-74%. The results suggested that the decontamination of molds by irradiation, before production of aflatoxin B1, is the most acceptable method in the preservation of peanut.

Effects of using different host plants on the detected biodiversity of arbuscular mycorrhizal fungi from an agroecosystem Rev. Bras. Bot.

The influence of peanut (Arachis hypogaea L.), sorghum (Sorghum bicolor (L.) Moench) and maize (Zea mays L.) on the development and diversity of arbuscular mycorrhizal fungi (AMF) from an agrosystem was investigated. Soil collected from an agricultural field where maize had been grown was inserted into sowing holes, under the seeds of peanut, sorghum and maize those were subsequently grown in sterilised quartz sand separately in plastic boxes for five months. After this period, collections of roots and rhizospheric soil were made to evaluate the percentages of root colonization (RC), number of spores (NS) and species of AMF. The genus Arachis (Fabaceae) native to South America, contains 80 species divided into nine sections, three of which contain species of special economic importance such as the cultivated peanut (Arachis hypogaea), belonging to the section Arachis, and some perennial forage species from sections Caulorrhizae and Rhizomatosae. We used microsatellite markers to assay genetic variability among 77 accessions of four species from section Rhizomatosae, the diploid Arachis burkartii (2n = 2x = 20) and the tetraploid Arachis glabrata, Arachis pseudovillosa and Arachis nitida (2n = 4x = 40). Genetic relationships among Arachis species based on AFLP Genet. Mol. Biol.

Amplified Fragment Length Polymorphism (AFLP) was used to establish the genetic relationships among 20 species from seven of the nine sections of genus Arachis. The level of polymorphism among nine accessions of the cultivated peanut, A. hypogaea L., was also evaluated. Three combinations of primers were used to amplify the AFLPs. The fragments were separated in 6% denaturing acrylamide gels. A total of 408 fragments were analyzed. An average of 135.3 fragments per primer combination were scored, and the largest number of fragments was 169 using primer combination Eco RI - ACC / Mse I - CTG, while the lowest was 108, with Eco RI - ACT / Mse I - CTT. We use a new partial-equilibrium, multi-market international model to analyze trade and agricultural policies affecting markets for peanut/groundnut products. The model covers four goods in thirteen countries/regions, including a large set of developing countries. Welfare is evaluated by looking at consumers' equivalent variation, quasi-profits in farming, quasi-profits in crushing, and taxpayers' revenues and outlays implied by distortions. We calibrate the model on recent historical data and current policy information. We analyze several groundnut trade liberalization scenarios in deviation from the recent historical baseline. The genus Arachis currently comprises 69 described species, some of which have potential and real value as human and animal foods. These Arachis species have been collected and maintained in germplasm banks to provide material that can be used as sources of genes in breeding programs and for the selection of new cultivars.
